Facility:

Sequencing of PCR
products, plasmid, phage and lambda DNA. We utilize ABI Prismtm
377 DNA Sequencer platform with BigDyetm
v.3.1 chemistry.

The sequences can
be obtained using custom primers (provided by clients) or our standard
primers (provided at no extra charge).

pGEM M13 sequencing
is used as a positive control with every run.

Custom primer design
for sequence walking.

Sequence assembly.
How
many nucleotides can be read from one reaction?
-
Sequence lengths
of greater than 650 bp are expected from high quality DNA templates.

The length of the
DNA sequence is highly dependent on the quality and accurate quantification
of the template.
Cost
of Services

For educational
institutions basic sequencing is provided at a cost of $15 per reaction
and $25 per reaction for both a miniprep and sequence. Industrial prices
are higher.

Turnaround
Time
Turn around
time is usually 2-3 days. If the time is greater, you will be notified.
Sample
Preparation and Shipment
Plasmid/Lambda/Phage
Samples:
-
The templates can
be sent either lyophylized (dry) or in WATER only (no salts please) on
ice.
-
A quantity of 0.5
ug per reaction is required at a concentration of 0.2 ug/ulor greater.
-
Please include
the type of vector, the size of the insert and an
accurate concentration.
PCR Samples:
-
The templates can
be sent either lyophylized (dry) or in WATER only (no salts please) on
ice.
-
A quantity of 20
ng per 100 bases in length of template at a concentration of 20 ng/ul or
greater.
-
Please state that
template is a PCR product, the length of the template and an accurate concentration.
-
Gel diagram is
helpful.
Primers:
-
Sequencing can
be obtained with either your custom primers or our standard primers
-
Primers must be
in water at a concentration of 1pmol/ul or greater or dry with known molar
value.
-
Please include
the concentration of your primers.
-
We can provide
most standard primers for sequencing vectors (M13, T7, T3, SP6, SK, KS).
Shipping:
-
Samples can be
mailed directly to the CSU Northridge DNA Sequencing Facility (see
contact information).
-
Samples can be
sent by standard mail or through a delivery service (UPS, FedEx, etc.)
depending on your needs.

Be sure to pack
your samples carefully!
-
Do not send samples
in single thickness envelopes - the US Postal Service is VERY hard on mail.
-
A padded envelope,
or a box is recommended as well as wrapping your tubes in Parafilm.
Data
-
Your sequence data
can be transferred electronically (both text and or electropherograms).
-
If you do not have
software for interpreting electropherogram files, a free version of EditView
(for Mac platform) can be downloaded from the
Apllied Biosystems website
(download starts automatically) PC platform based software
packages include Chromas, GeneTool and others.
-
Hard copies of
the electropherograms will be mailed upon your request.
-
If you wish a hard
copy of the text and/or electropherogram data files please include a zip
disk.
